番茄是世界范围内的重要蔬菜作物,同时也因为其较小的基因组,完整的遗传图谱以及较短的生命周期被作为研究果实生长成熟的模式植物。对于番茄果实成熟的研究不仅能帮助了解果实成熟的机制,也对提高番茄果实品质,以及其他肉质果实品质具有重要的作用。近年来,番茄的果实成熟被越来越广泛深入地研究,果实成熟的调控机制也越来越清晰。 最新研究显示,DNA甲基化可能是转录因子和植物激素之外的第三个最重要的调节果实成熟的因子。然而,DNA甲基化参与调控果实成熟的机制则缺乏相应的深入研究。

我们将以番茄为研究对象,结合遗传学,生物信息学,分子生物学等多种研究手段来研究DNA甲基化对果实成熟的调控机制。相比较最常用的模式植物拟南芥,在番茄中进行DNA甲基化机制的研究有其优势及独特性。之前的研究表明,DNA去甲基化在拟南芥中对胚胎中基因印记十分重要,然而在成熟的植物体中,DNA去甲基化主要作用于转座子及基因间区域,很少影响编码基因的表达。在番茄的果实中,已有证据显示,DNA去甲基化有可能参与了大量的与果实成熟相关的基因表达。因此,以番茄为研究对象,能揭示DNA去甲基化对植物基因表达和生长发育的重要功能。
